Project Profile: This project aims to link with enhancing Raman spectrum and dark field scattering spectrum, to establish a kind of SERS (Surface Enhanced Raman Scattering) detection and analysis method based on metal oxide single particle and high throughput, which is used for bacterium typing actual imaging and optical spectrum detection. Develop different type metal oxide single nanometer particle synthetise strategy and interface organic molecular modification technology. Establish multi-targeted metal oxide nanometer particle probe modified by different kind of organic molecular. Upon developing metal substrate microprocessing technology, establish microarray probe metal substrate modified by different kind of organic molecular. Upon identifying specificity of target molecular, establish three-layer sandwich structure of metal oxide single nanometer particle - target molecular - metal substrate. Under the effect of target molecular, the metal oxide nanometer particle will be anchored in specified area of metal substrate. The metal oxide nanometer particle will arouse metal substrate surface plasma resonance. High local electromagnetic field hotpoint will be formed at juncture area. And accordingly, increase Raman signal strength. Linked with reflect dark field microtechnique, establish real time dark field imaging and SERS optical spectrum detection device, and accordingly, realize high sensitive, high throughput SERS detection and dark field imaging for bacterium typing. Provide new technology and analysis method for realizing earlier diagnose and screening bacterial infection, and for developing family type portable instrument.
